Modafinil

I dont like coffee, that's fine with me, the only thing that always bugged me is that peoples drink coffee to wake up. I don't have much alternative, i tried some xenidrine, but that's not really good for the health, so I just stopped that.

Until, Recently I saw that new drug ! I did a cut and paste of the description from google cache since the original page was down. I often feel very sleepy at work, everywhere, when i go out etc, I believe this is a physical problem maybe. (And a part of the reason why im lazy since im tired). The Text say the generic drug should be out in 2006, and that Alertec in Canada is cheaper. I think I will try some really soon or maybe I just wait the generic drug....

Modafinil: "...modafinil ('Provigil', 'Alertec', 'Vigicer', 'Modalert', etc) is a memory-improving and mood-brightening psychostimulant. It enhances wakefulness and vigilance, but its pharmacological profile is notably different from the amphetamines, methylphenidate (Ritalin) or cocaine. Modafinil is less likely to cause jitteriness, anxiety, or excess locomotor activity - or lead to a hypersomnolent 'rebound effect' - than traditional stimulants. Subjectively, it feels smoother and cleaner than the amphetamines too. The normal elimination half-life of modafinil in humans is between 12 - 15 hours. So it's worth fine-tuning one's dosage schedule accordingly.

Current research suggests modafinil, like its older and better-tested analogue adrafinil, is a safe, effective and well-tolerated agent. It is long-acting and doesn't tend to cause peripheral sympathetic stimulation. Yet its CNS action isn't fully understood. Modafinil induces wakefulness in part by its action in the anterior hypothalamus. Its dopamine-releasing action in the nucleus accumbens is weak and dose-dependent; the likelihood of a euphoric response ('abuse potential'), dose-escalation and tolerance is thus apparently small. Modafinil has central alpha 1-adrenergic agonist effects i.e. it directly stimulates the receptors. Modafinil inhibits the reuptake of noradrenaline by the noradrenergic terminals on sleep-promoting neurons of ventrolateral preoptic nucleus (VLPO). More significant, perhaps, is its ability to increase excitatory glutamatergic transmission. This reduces local GABAergic transmission, thereby diminishing GABA(A) receptor signalling on the mesolimbic dopamine terminals.

Modafinil is proving clinically useful in the treatment of narcolepsy, a neurological disorder marked by uncontrollable attacks of daytime sleepiness. Narcolepsy is caused by dysfunction of a family of wakefulness-promoting and sleep-suppressing peptides, the orexins. Orexin neurons are activated by modafinil. Orexinergic neurons are found exclusively in the lateral hypothalamic area, but their fibers project to the entire central nervous system. Genetically modified orexin-knockout animals offer a model of human narcolepsy. Selective orexin receptor agonists of the future may prove useful both to narcoleptics and the population at large.

Experimentally, modafinil is also used in the treatment of Alzheimer's disease, depression, attention-deficit disorder, myotonic dystrophy, multiple sclerosis-induced fatigue, post-anaesthesia grogginess, cognitive impairment in schizophrenia, spasticity associated with cerebral palsy, age-related memory decline, idiopathic hypersomnia, jet-lag, and everyday cat-napping. Depressives who feel sleepy and fatigued on SSRIs can augment their regimen with modafinil. In September 2003, an advisory panel to the FDA endorsed its use for treating shift work sleep disorder and obstructive sleep apnea.

The US military are interested in modafinil too.

Modafanil is marketed as 'Alertec" in Canada - and over the Net. 'Alertec' is less expensive than 'Provigil'. Cheap generic modafinil should be available from 2006. But Cephalon is vigorously litigating to defend its patents.

In March 2005, Cephalon filed a New Drug Application (NDA) with the FDA for "Nuvigil" (r-modafinil, armodafinil) - a single isomer formulation of modafinil. Nuvigil will be marketed aggressively to offset the loss of revenue from Provigil.

Modafinil is increasingly used as a 'lifestyle drug' - a lucrative 'off-label' market its makers have not been unduly keen to discourage. Some prescribing physicians have reportedly been surprised at a previously hidden epidemic of narcolepsy among hard-working young professionals attending their surgeries.

Prudence, however, should be exercised in drastically curtailing one's sleep. Prolonged sleeplessness weakens immune function. Animals tortured in sleep-deprivation experiments eventually die from massive bacterial infections of the blood..."

I found a good way, when i have time to "spare" at work and I dont know what to check on the web to entertain myself, Im randomly checking other blogs. Yesterday was easy, first one was good and second one too. Today took me a while, I got a lots of others languages blogs and strange stuff. Finally a "standard" one:

I should put pictures in my blog, Every blog I visit that I like has nice ones. Actually some facelift too would help.

Actually knowing myself, its more interesting reading women blog. (Especially those with some pictures of themself). I have to remember add my picture on my blog on of these day. Like tonight =P

Microsoft is supposed to release Virtual Earth with better pictures and building with satellite picture of 45 degree, should be good.

Keira Knightley

Yep, she is hot like a lots of women anyway, but geesh these eyes.
I was checking 3-4 pictures from http://www.dominomovie.com and i find her eyes, that look, that "glance", whatever the name soooo special.

This say, Im a realist person. I rather spend time trying to meet another women than her, since my chance with her are from 0.00000000001 to 0.0000000002 (thats 1 in 2 billions or so :)

I went to the Québec National Celebration (St-Jean Baptiste) yesteday at the Parc Maisonneuve, and god it was boring, the music was not loud enough (its rare for a show). The song choice... god horrible, all slow song without rhythm(sp?), no beat. Overall boring except some nice looking women left and right sometime.

Cutting the show crap i saw recently (Except the Boxe at the Centre Bell was really cool but i was drunk like a pig, stupid me).

The PSP god a real exploit for the version 1.5, I can now play all emulators on my PSP ! Thats awesome to play Nintendo game, Super Snes, Genesis (name it you have them) on it ! That with special homebrew programs (Like doom, etc.) See http://psphacker.com for more detail about it (Be sure to have a adblock loaded on your firefox, because the page is ads, ads, ads but really good info overall).

Thats it for now, a lots to say, a lots probably not interesting anyway, maybe i should write movie review, or review on rottentomatoes, since i watch a tons of them... Hmmm because I wont talk about all the beers i went to take, there will be 50 boring posts of where I was taking beers.

Oh thats remember me, If you ever need a Passport in Canada, for god sake. Fill your demand on the Internet web site. I did that with ePass whatever the fed gov account thing. Boring ot do but worth it, rather than spending 2 hours at the building waiting. I got in a special line, took me 20 min from the elevator to the passport building to the way back to my car outside ;) Next best thing. The passport is usually 3weeks-2months+, Mine was home 6 days after, yep Friday to Thursday. I already got it impressive.

Beers, Summer and Beers

Beer Festival is this week in Montréal, This is my destination after work :) A good timing, its been raining for like 3 weeks and for today and yesterday we got a big shiny SUN, everyone is excited around. Its hot outside, women remove more clothes you know the patern.

PSP vers 1.5+ still don't run homebrew and emulators, but we getting so close. I cant wait to play punchout on my PSP. There is a tons of rumor about loaders etc. Hopefully within the next 2 weeks something will be out.

So many thing to say, so many thing that I will probably not say on a blog =P Personal feeling on a lots of stuff etc. So the blog will stay for events and geeks tech news related, games etc I guess.

Why nothing about personal feeling or the way the world work ? Because:
Women Are Evil :)

I should explain my theory someday !
